Here is the Wikipedia link and here is Rokuro’s, but it's a little sparse and doesn't summarize episode 9, so I'll do a brief summary as well.
While not much is known on Rokuro, we do know that he has served both Yukimura and his father, likely for most of if not his entire life--his family as well has a history of serving the Sanada clan. He is seen always serving Yukimura and is constantly at his side.
The events of the anime (and manga) follow Yukimura’s attempt to gather 10 “braves” to serve him and fight for him which will allow him to tame the resident chaos goddess. It’s a very serious plight, I promise.
These 10 braves will each embody an element, and thus begins his journey to gather everyone he needs while navigating the Sengoku era in war-torn Japan. The first they find is Sasuke, who acts as ‘leader’ of the braves, and Ana as well—following that, Isanami and Saizou stumble upon Yukimura through a series of unfortunate events. Yukimura takes them in, of course, designating them a part of his braves.
It is with the arrival of Isanami that things truly begin to happen. Isanami carries with her the kushi mitama, and within her is the potential to throw Japan into chaos. It is because of this that she is hunted. Meanwhile, Yukimura must deal with the struggles of retaining his territory in a Japan filled to the brim with hungry unifiers-to-be (Ieyasu Tokugawa being the biggest offender, though Masamune Date is certainly not far behind).
Thus follows both shenanigans and serious moments of fighting to defend what they believe in.
Of rather serious importance is episode 9, which follows right after Yukimura has finally finished assembling his 10 braves and has shared with Saizou his intentions. Ana betrays the group then, as she had been working for another master the entire time (and everyone had been far too blinded by her cleavage to notice otherwise). Ana attacks Rokuro in his sleep, aiming to steal away his right eye. Rokuro’s right eye has the ability to remember anything it records—in this way, it held Yukimura’s plans and all of the information that her master wanted. She managed to pin him, but as she was trying to cut out his eye, Rokuro lunged forward so that she would stab his eye entire, for as he said, he would rather lose it than allow her to steal it away.
Ana also grievously harms Sasuke when he crashes in to save Rokuro and try to stop her, and proceeds to flee the company.
Yukimura is quick to clean and bandage uphis wife his attendant, and Rokuro is left to rest and recover while the others attempt to pick up the pieces of what had been a happy family of sorts.
And then Rokuro ends up the island. Oops.
Personality:
Rokuro is the epitome of a loyal retainer—to an extent.
He has his special habits; he grumbles at his lord for his abuse of alcohol and love of women, and he is not at all against scolding Yukimura when he does things that he disapproves of. Nor is he at all against trying to make his lord see reason in various ways—he brings out a game in order to try to show Yukimura what he is missing, though as per usual Yukimura is just a few steps ahead of him already.
Still, it is his role to serve Yukimura utterly and fully—in that way, he is akin to the “slave” that was meant to follow behind their “master” to whisper into their ear “remember that thou art but mortal”. Even if Yukimura is sharper than he acts or appears and even if Yukimura is on top of everything 90% of the time, Rokuro will continue to perform that role, for it is a sacred role that he takes utterly seriously.
And of course, he is unbelievably loyal. What Yukimura says shall be done, and without hesitation. When it comes to the larger things, there is no room for hesitation, and to be honest, Rokuro would not think of doing so. He might grumble about the small, ultimately unimportant things, but he quietly bows to all of importance that Yukimura believes in, because he does have faith in his lord.
Beyond that, Rokuro is quiet and lineface a good majority of the time. He tends to take things in stride, and really, only Yukimura is good at consistently ruffling his feathers (Yukimura probably does it because he thinks it’s amusing). From protecting his lord from intruders to being attacked in the middle of the night, he tends to take things as they come and deal with them without panic or over-emotional displays. (It could almost be concerning how little he reacts to Ana straddling him in the middle of the night--)
But there are times when he isn’t lineface. He dislikes it when people get too close to Yukimura, it’s true, especially if he doesn’t trust them fully. There is a moment in which he hastily slaps away Ana after she hugs Yukimura from behind. He prefers the quiet and prefers to stick to the background, acting as the wallflower of the scene, but when it comes to Yukimura he does not ever hesitate to act.
He’s usually calm and pretty chill, suiting his position as the “water” of the elements Yukimura is collecting, but again, there are things that make him react quickly. Threats to Yukimura are dealt with quickly and succinctly (and again without a whole lot of unneeded emotions).
He isn’t as lineface as he would like to be, of course. While he does tend towards being distant from the other braves, the impartial observer as they grow closer and bond, he is nonetheless grateful to them for their service to Yukimura, even serving all of them at the hot springs, as his lord had wanted to go. He merely has his priorities, of which Yukimura is the highest and most unshakeable; to serve Yukimura well and faithfully is the fastest way to get on his good side. And, on the flip side, to harm Yukimura is to receive a swift death.
Rokuro does have his fair share of issues, however. With the loss of his eye comes the feeling that he has failed his lord. Yukimura told him to protect and keep careful hold of his right eye, and Rokuro failed to do so, though he did keep it from falling into anyone else’s hands by harming himself on Ana’s blade. Nonetheless, he still apologizes multiple times for failing Yukimura, for not only has he lost his eye when Yukimura told him to keep it safe, he has lost one of the ways in which he served his lord. He cannot help the feeling that he has failed Yukimura utterly, and for that he is crushed and beyond sorry.
And thus Rokuro is the quiet observer, the untrusting, cynical retainer who keeps constant watch. Serving Yukimura is not his goal, it is his life.
Abilities/Strengths:
Rokuro is one of the braves, a ninja capable of combat. He is capable both of hand-to-hand combat as we see in an early scene when he is tied up with Yukimura and defends them both, and also can use two triangular blades to fight. He also uses said blades to amplify and fight with ultrasonic soundwaves, though he will be incapable of doing so in Arcanarum.
Once upon a time he could also “absorb” letters and words from manuscripts with his right eye and reduplicate them perfectly, as if with a photographic memory, but he lost the eye and with it that ability.
Otherwise, he is an excellent wife.
Weaknesses:
Yukimura is most certainly Rokuro’s first and most glaring weakness, as there is nothing Rokuro would not do for the man. He could be manipulated into anything were Yukimura involved, and he would do anything with but a word from his lord. Beyond that, he is most certainly not the strongest of the braves in terms of fighting; he acts more as a retainer than a fighter, though he is most certainly capable and serves as Yukimura’s bodyguard.
Also he’s kind of a lineface fun sucker which is totally a weakness.

Story Character: Lakhesis of the Moirae
Plans: Rokuro is known as Rokuro of the Fates; he had an eye that would allow him to remember everything he saw. In a way, that makes him fit the role of the one who measured men's lives by the length of their string before it is cut. However, he cannot see into the future and, in fact, ends up making a number of nearly mortal mistakes, and he is of course far too human to ever judge peoples' lives, lineface as he is. As for plans in game, the status of the fates as impartial goddesses will likely affect his ability to retain his compassion and humanity, even amongst his lords and friends, which would be fascinating to deal with.

He was lying in grass.
That was the first thought that came to him as he resurfaced from unconsciousness. Why was he lying in grass...? And in such a peaceful atmosphere, too. He could hear bird chirps in the fuzzy area between consciousness and unconsciousness that he now rested within, fingers twitching in the grass that brushed them.
And then Rokuro moved and it was blinding pain.
But it was also remembrances.
He remembered--betrayal and frustration and fighting and his own failures--
And so it was that despite the pain, he bolted up to a sitting position, fingers grasping for the wound in his shoulder as he flinched automatically, breath coming shorter with the pain that shot through it. But that didn’t matter.
What mattered was that Yukimura was not there, and Rokuro was no longer in Ueda castle.
But that...that was impossible. He could not have ended up...here.
But here he was, despite the impossibility of it all, and so, bleeding and his now ruined eye tightly shut, Rokuro managed to stagger to his feet. He did not know where the other Braves were, but that was of secondary importance; most importantly, he did not know where Sanada Yukimura was. He did not know where he himself was, but most of all…He did not know where his lord was.
He had to find him.
He knew he was in no shape to deal with this situation--but he had to find his lord, had to apologize, had to protect him and make up for his failures. There was no option; it was not a choice. It was a fact, and Rokuro would not fail his lord again.
And so he put the strangeness of the scenery around him aside and began his search, leaving a trail of dripped blood behind him.
